摘 要： 利用单因素法对枯草芽孢杆菌发酵产碱性蛋白酶的培养基成分进行优化，确定发酵培养基各成分最适宜配比。最后确定了该菌株的最适发酵培养基配方为(g/ 100 mL) :棉籽饼粉3.0 ,酵母浸粉1.75 ,麦芽糊精10 ,柠檬酸钠0.13 ,CaCl2 0.13 。K2HPO4 1.0 最适摇瓶发酵条件为:接种量2 % ,装液量50 mL/ 250 mL ,摇床转速200 r/ min ,34 ℃,发酵54 h ,碱性蛋白酶的发酵单位可达18.8u/ mL。此外,还进行了5 L 罐放大实验,在5 L 罐所确定的最适工艺条件下,发酵单位可达 520.2 u/ mL 。
